         Bst3.2 DNA聚合酶经酶电子重构架和进化筛选(in silico Design & invitro Evolution),将Bst2.0和Bst3.0特性融合,保留了3.0的快速扩增能力和增强的内置RT活性,融合了2.0的扩增特异性佳和dUTP使用能力(允许使用50%的dUTP来代替dTTP)。该酶具有5’-3’的DNA聚合酶活性和强烈的链置换活性,但其5’-3’和3’-5’外切酶活性缺失。
         Bst3.2 DNA聚合酶用于RT-LAMP实验,尽管Bst3.2 DNA聚合酶本身具有良好内置的RT活性,但在低Copy样本的RT-LAMP中表现出反应速度滞后,因此在RT-LAMP反应时建议搭配耐热逆转录酶Max65 RTase(HaiGene D0303-02),该组合方案下Bst3.2 DNA聚合酶的使用性能等同于HaiGene的Bst4.0 DNA/RNA聚合酶,该酶是Bst4.0 DNA/RNA聚合酶的核心组分。

Bst3.2 DNA聚合酶对RNA模板的扩增能力展示:分别用Bst 2.0(0.32U/μl)、3.2(0.32U/μl)、3.2(0.32U/μl)+Max65 RTase(2U/μl),以A549细胞Total RNA为模板,进行RT-LAMP扩增。
